EGFR expression IHC analyses
• Histopathologic studies are retrospective
• Material selected based on tissue availability
• Several antibodies for IHC analysis
• Lack of uniform criteria for defining positive EGFR expression
• Prospective studies requested to assess whether EGFR expression predicts treatment outcome

Predictive Biomarker Studies in Tumor and Skin Tissue Samples of Patients With
Recurrent H&NC Neck Treated With Erlotinib
Aglunik et al J Clin Oncol 25, 2007
• 37 samples, response to therapy 2 / 4 in high EGFR gene copy number tumors VS 4 / 27 (15%) in low gene copy number tumors
• High EGFR gene copy in tumor specimens may predict which patients have an increased likelihood of response to erlotinib
• With treatment, a decrease in p-EGFR, p-NFB, and p27 correlated with increased TTP, OS, or both TTP and OS, respectively (P .04)
• Decreased p-EGFR level in skin biopsies during therapy may represent a potential surrogate marker for improved clinical outcome.
Milas L. IJROBP, 67: 568–572, 2007
Ratones A431 xenoingertos fueron tratados con:
• RT sola (14 fracciones)
• RT + cetuximab x3
• RT + cetuximab x3 cetuximab x 3 (mantenimiento)
Control of A431 tumor xenografts by local irradiation combined with C225
Milas L. IJROBP, 67: 568–572, 2007
Resistencia adquirida a cetuximab
